Quick Reference:
One By One: In the client/lead record, click on the headset icon next to their number to add that number to the DNC list in your Radius account. Optionally add a reason. Submit.
In Bulk: Settings> Phone Dialer> Do Not Call List> Import a csv file that lists Phone Number as the header label and the phone numbers on that list (and optionally a column for Reason)

To add someone to the Do Not Call list, simply click over the headset icon and click on it.
It will then prompt you to add that number to the Do Not Call list.
You can optionally add a reason. Most of the time it's because they're on the Do Not Call list or because they asked for it, but otherwise you, You could put in a reason like prefers text. That way, if they really need a call, you understand that it's prefers text rather than, you know, that they're on the DNC.
To remove them from DNC, simply click on the little no sign and then you can remove that number.
But if you have it on the DNC and you try to call them anyway, it will tell you it's unable to place the call and the reason why.
To do this in bulk, you will go down to Settings> Phone Dialer (Edit)> Do Not Call List.
You will add the numbers in bulk. You can also just add them one by one, but that's boring. Hit import do not call phone numbers. You will need to upload a CSV file for PHONE NUMBER being the column header label and, optionally, a column for REASON. Usually if you're doing a large import, it's because you've scrubbed against the do not call list for the leads that you've imported, which is helpful to know. You will upload the file by hitting browse. Add the file in and then hit import do not call phone numbers.
It will be queued for import. Now, if I look in my settings, phone dialer, do not call list. They are now added to the DNC list, along with the reason.
Important to note, you must name the field headers, PHONE NUMBER and REASON. If you just put in PHONE, it will not work.
You can delete a number from the list if you want to remove it from the account's do not call list.
